Its basic principle is to culture cells or tissues on a gas-permeable, porous, elastic artificial matrix and promote their growth and development through simulated physiological stimuli such as joint movement, pressure, and fluid shear. This simulation helps study cell behavior under specific physiological conditions and the mechanisms of disease onset and progression.
1. Main control box: the control center of the entire reactor, responsible for setting and controlling mechanical-environment parameters in the culture chamber. It simulates the pulsatile mechanical environment of cerebrospinal fluid by setting parameters such as pressure and frequency for the liquid in the connected silicone tubing.
2. Cell incubator: provides a suitable cell-growth environment, including constant temperature and humidity. It is generally positioned below the main control box for convenient operation and maintenance.
3. Culture chamber: located inside the cell incubator, this is where cells are grown and cultured. Silicone tubing and fittings connect it to the main control box so that mechanical parameters are accurately transmitted to the chamber. Each culture unit includes two scaffolds, one experimental and one blank control, and each dynamic culture unit can be installed, removed, and relocated as the experiment requires.
4. Silicone tubing and fittings: connect the main control box and culture chamber and ensure that the chamber's mechanical environment matches the parameters set by the controller. They offer an appropriate elastic modulus and good mechanical transmission and are non-cytotoxic and biocompatible.
